Dear Sir, How do I assume that for a given question when should I take Permutation and when to take Combination or both ??

If the order matters, it is a Permutation.
 
If the order does not matters, it is a combination.
 
Consider an example:
 
From a deck of 52 cards, we need to select 5 cards.
How many different combinations are there?
 
There are 52C5 different combinations are there.
 
Now consider the example of allotting 3 prizes to 3 students.
 
Suppose there are three students A, B and C
 
So we can assign first, second and third prizes in the following ways:
ABC, ACB, BCA, BAC, CAB, CBA
 
Thus, we can assign the prizes in 3P2 ways.
