What will happen if we use a negative test charge for testing the electric field ? what would be the direction of electric field?

If you would use the negative test charge, then the direction of the electric field would be opposite to that of the force experienced by the negative charge. If you will take that opposite effect into account, you should still get the same direction of the electric field as if you would have used a positive test charge.
