- In 1957 Skoog and Miller put forth the concept of hormonal control of organ formation.
- According to this, root and shoot differentiation is a function of the auxin-cytokinin ratio.
- Relatively higher concentration of cytokinin promotes shoot formation. So, if the medium used for tissue culture contains cytokinins in higher amounts as compared to auxins, the tissue differentiates to form the shoot or stem of the plant.
- Relatively higher auxin concentration favours root formation. So, if the medium used for tissue culture contains auxins in higher amounts as compared to cytokinins, the tissue differentiates to form the root of the plant.
