Capacitance C of parallel plate capacitor related to plate separation distance d as
 
C = ε ( A / d )  
 
where A is area of capacitor and ε is  electric permittivity of the medium between plates .
 
Hence if we know the capacitance C and area A , we can calculate the disstance d between plates.
 
Voltage across the plates will not help us to get the distance d between plates
