while calculating voltage difference do we take the positive or negative sign of the charge?

Potential difference is given as V = W/Q.
If the sign of the charge is either positive or it is not mentioned, then it has to be taken as positive.
If it is a negative charge, then it has to be taken as negative while calculating potential difference.
